TNF Superfamily Ligands

About 19 tumour necrosis factor (TNF) superfamily (TNFSF) ligands are currently identified, which include TNF-α, TNF-β, lymphotoxin-β (LT-β), CD27L, CD30L, CD40L, FasL, 4-1BBL, OX40L, TRAIL (TNF-related apoptosis-inducing ligand), LIGHT (homologous to lymphotoxins), RANKL (receptor activator of NF-κB ligand), TWEAK (TNF-related weak inducer of apoptosis), APRIL (a proliferation-inducing ligand), BAFF (B-cell activating factor), VEGI (vascular endothelial cell-growth inhibitor), ectodysplasin A (EDA)-A1, EDA-A2, and GITRL (glucocorticoid-induced TNFR family-related gene ligand). Structurally, TNF ligands are type II transmembrane proteins that share a common structural motif, the TNF homology domain (THD). The THD binds to a cysteine-rich domain (CRD) in the NH2 terminus of TNF receptors. The THD is responsible for the trimerization of TNF ligands and their binding to the trimerized receptor complex. Interactions between TNFSF ligands and TNFRSF receptors mediate signaling that controls the survival, proliferation, differentiation, and effector functions of both immune and non-immune cells. Noteworthy, TNFSF ligands, while regulating normal functions such as immune responses, haematopoiesis and morphogenesis, have also been implicated in tumorigenesis, transplant rejection, septic shock, viral replication, bone resorption, rheumatoid arthritis and diabetes.
