MCE Live & Dead Bacterial Viability and Counting Kit is based on dual-fluorescence staining technology, which can effectively differentiate live bacteria from dead bacteria and enable quantitative detection using flow cytometry. It can also be used for quantitative analysis of various bacterial mixtures.

The detection principle is as follows: the green nucleic acid dye, which has membrane permeability, can easily penetrate both Gram-negative and Gram-positive bacteria, emitting a bright green fluorescence signal, and can stain both live and dead bacteria. The non-membrane-permeable red dye, PI, can only stain dead bacteria with compromised cell membranes, emitting red fluorescence. With the appropriate mixture of the green nucleic acid dye and PI dye, bacteria with intact cell membranes emit bright green fluorescence, whereas bacteria with damaged cell membranes exhibit significant reduction in green fluorescence and emit stronger red fluorescence. By adding an appropriate amount of detection microsphere before analyzing the sample on a flow cytometer, dual-fluorescence staining flow cytometry can be used to detect the concentration of both live and dead bacteria. Both the green nucleic acid dye and propidium iodide dye are efficiently excited by the 488 nmspectral line of the argon-ion laser found in many flow cytometers, with the fluorescence signals detected in the green and red channels, respectively. Tthe background remains virtually non-fluorescent.

MCE Live & Dead Bacterial Viability and Counting Kit has been tested at most bacteria, such as Escherichia coli, Bacillus cereus, Bacillus subtilis, Clostridium perfringens, Mycobacterium phlei, Pseudomonas aeruginosa, Klebsiella pneumonia, Micrococcus luteus, Pseudomonas syringae, Staphylococcus aureus, and Streptococcus pyogenes. This kit (100 T) contains sufficient material to perform approximately 100 individual assays by flow cytometry.

Protocol

1. Preparation of bacterial samples

Resuspend bacteria in sterile PBS or saline and adjust to a concentration suitable for flow cytometry. Prepare a viable-bacteria control and a heat- or alcohol-killed control when possible.

2. Fluorescent staining

1. Equilibrate fluorescent dyes to room temperature and briefly centrifuge before use.

2. To 1 mL bacterial suspension, add 2 μL Green Nucleic Acid Staining Solution (3.34 mM) and 2 μL PI Staining Solution (20 mM), and mix gently.

3. Incubate for approximately 15 min at room temperature protected from light.

Note: A 1 : 1 mixture of the two dyes is generally suitable for most bacterial staining assays. Optimize the ratio according to bacterial species and Gram characteristics when necessary.

3. Counting microspheres

Thoroughly mix Detection Microsphere immediately before use. Add 20 μL Detection Microsphere to each stained sample, mix gently, and analyze immediately by flow cytometry.

4. Flow-cytometric analysis

Green fluorescence primarily identifies viable bacteria, whereas PI red fluorescence identifies membrane-compromised dead bacteria. Calculate total bacterial concentration and viable fraction from bacterial and microsphere event counts together with the known microsphere concentration.

Note: Do not freeze Detection Microsphere. Protect all fluorescent dyes from light.

Attention

1. The fluorescent dye should be equilibrated to room temperature before use. It is recommended to briefly centrifuge before aspirating.

2. Fluorescent dyes are susceptible to quenching, Minimize light exposure during handling and storage to delay quenching.

3. Cell type and Gram characteristics may affect the intensity of red fluorescence generated by dead bacteria.

4. PI is harmful to humans, Exercise appropriate safety precautions during handling.

5. A 1:1 mixture ratio of the two dyes is generally compatible with most bacterial staining analyses. The ratio may be adjusted empirically to optimize staining performance for specific applications.
