Description & Advantages

MCE BCA Protein Assay Kit is a colorimetric assay based on the bicinchoninic acid (BCA) reaction principle for quantitative protein determination.

The detection principle is as follows: under alkaline conditions, proteins reduce Cu2+ to Cu+, and the resulting Cu+ forms a purple-colored complex with BCA molecules. This complex exhibits a strong absorbance at 562 nm, with the absorbance showing a linear correlation with the protein concentration in the sample. This product enables accurate quantification of protein concentrations in the range of 0-2000 µg/mL.

1. Preparation of BSA Standards

Prepare the protein standard solution using the supplied BSA and generate a series of standards according to the expected sample concentration range. A fresh standard curve is recommended for each protein assay.

2. Preparation of BCA Working Reagent

Prepare the required amount of BCA working reagent by mixing Reagent A and Reagent B at a volume ratio of 50 : 1. Mix thoroughly before use. Prepare approximately 5%-10% excess working reagent according to the number of wells.

3. 96-Well Plate Method

BCA working reagent 200 μL

1. Add BSA standards and protein samples to a 96-well plate. Replicate wells are recommended.

2. Add 200 μL freshly prepared BCA working reagent to each well and mix gently.

3. Incubate at 37°C for 30 min.

4. Cool to room temperature and measure absorbance at 562 nm.

5. Plot BSA concentration versus A562, calculate sample protein concentration from the standard curve, and multiply by the corresponding dilution factor.

Note: The quantitative range of the kit is 0-2000 μg/mL. If samples contain high concentrations of interfering substances, dilution or dialysis is recommended.

Attention

1. It is recommended to prepare a new standard curve for each protein assay to ensure the accuracy and reproducibility of the results.

2. If the sample contains high concentrations of interfering substances, dilution or dialysis is recommended. If accurate results still cannot be obtained, alternative protein quantification methods—such as the Bradford Protein Assay Kit (Cat. No.: HY-K2001)—can be used.
