Verification Images

  • Experimental Validation Results for ELMO1 Antibody (YA2243)
Western blot analysis of extracts from Jurkat (lane 2(20μg), C6 (lane 3(20μg), NIH/3T3 (lane 4(20μg), using ELMO1 Antibody. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.
  • Experimental Validation Results for ELMO1 Antibody (YA2243)
Immunocytochemistry analysis of Hela cells labeling ELMO1 with ELMO1 Antibody (HY-P82498) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with ELMO1 Antibody (HY-P82498) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L(HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

Background

  • Function

ELMO1 is involved in cytoskeletal rearrangements required for phagocytosis of apoptotic cells and cell motility. Acts in association with DOCK1 and CRK. Was initially proposed to be required in complex with DOCK1 to activate Rac Rho small GTPases. May enhance the guanine nucleotide exchange factor (GEF) activity of DOCK1

  • Subcellular Localization

    Cytoplasm; Cell membrane

  • Expression

Tissue_specificity:Widely expressed, with a higher expression in the spleen and placenta

  • Isoforms & Post-Translational Modification

    Q92556 has 3 isomers: Q92556-1: 83829 Da (predicted); Q92556-2: 28742 Da (predicted); Q92556-3: 49915 Da (predicted).
    Phosphorylated by HCK

  • Subunit

Interacts with ADGRB1 (By similarity). Interacts directly with the SH3-domain of DOCK1 via its SH3-binding site. Part of a complex with DOCK1 and RAC1. Part of a complex with DOCK1 and CRK isoform CRK-II. Interacts with PLEKHG6. Interacts with HCK (via SH3 domain). Interacts with ADGRB3 (PubMed:24567399). Interacts with DOCK5 (By similarity)
