MCE Antibody Labeling Kit-VF 488 can efficiently and covalently attach the VF 488 dye to antibodies. The antibody labeling process is straightforward and rapid, eliminating the need for purification post-labeling. Each antibody molecule binds covalently with 4-6 dye molecules, allowing the labeled antibody to be utilized in multifluorescent staining experiments without any inter-antibody dye transfer issues. Moreover, the ultrafiltration tube provided in the kit allows for antibody sample concentration or buffer exchange. Different kit sizes are available to accommodate the reagents necessary for labeling various quantities of antibodies.

Protocol
General Protocol
1. Antibody preparation
The antibody should preferably be in a buffer free of excessive primary amines or high concentrations of carrier proteins. If the antibody is too dilute or the buffer is unsuitable for labeling, use the supplied 10 kDa MWCO Ultrafiltration Tube for concentration or buffer exchange.
Note: Before labeling, avoid high levels of Tris, glycine, BSA, gelatin, or other components that may interfere with the reaction of the NHS ester dye with primary amines on the antibody.

3. VF 488 labeling reaction
1) Transfer the antibody sample to a suitable tube and select the corresponding VF 488 Dye vial according to the antibody amount.
2) Add Reaction Buffer (10×) to the antibody to establish the appropriate mildly alkaline labeling conditions and mix gently.
3) Transfer the antibody solution into the VF 488 Dye vial and mix thoroughly but gently so that the dye is fully dissolved and contacts the antibody.
4) Incubate for approximately 30 min at room temperature protected from light. No additional optimization of the dye-to-antibody ratio is required.
5) After labeling, add the corresponding amount of Antibody Storage Buffer and mix gently. No additional purification is required before use in downstream applications such as immunofluorescence or flow cytometry.
Note: VF 488 Dye is light-sensitive. Protect the reaction and labeled antibody from light. For long-term storage, aliquot according to antibody stability and avoid repeated freeze-thaw cycles.
