MCE Blasticidin S, Sterile (10 mg/mL) is a filtered and sterilized antibiotic solution that can be used directly in cell culture. Blasticidin S is a peptidyl nucleoside antibiotic isolated from Streptomyces griseochromogenes. It acts by blocking hydrolysis of peptidyl-tRNA induced by release factors and inhibits peptide bond formation. It can be used as a selection agent in both mammalian cells and bacterial cells.

Resistance to Blasticidin S is conferred by bsr and BSD. Therefore, Blasticidin S is a popular and effective antibiotic for selecting vectors bearing the bsr or BSD gene in mammalian cell lines. The recommended working concentration ranges from 1 to 20 μg/mL depending on the cell line and 50-100 μg/mL for bacterial selection. For bacterial selection purposes, the salt content of the LB medium must remain low (<90 mM) and the pH should not exceed 7.0 to maintain the activity of Basticidin S.

Protocol

The Blasticidin S solution can be used according to the following methods:

1. Add Blasticidin S solution in proportion to the sterile cell culture solution, mix well before use.

2. Add Blasticidin S solution in proportion to the cell culture solution (the cell culture medium contains bacteria), filter and sterilize.
