Description & Advantages

MCE Mouse Liver Ductal (Differentiation) Organoid Kit includes a basic culture medium for Mouse Liver Ductal (Differentiation) organoids, as well as culture supplements, and is designed for the establishment and long-term maintenance of mouse liver ductal organoids (mLDs) derived from adult stem cells.

The self-renewal of the ductal epithelium is sustained by the proliferation of stem cells and their progenitors residing in the liver. The mouse liver ductal organoids (mLDs) recapitulate key features of the native ductal epithelium, including tissue architecture, cellular composition, and self-renewal dynamics. As such, they represent a powerful in vitro model for advancing studies on mouse liver development and disease. Under appropriate differentiation conditions, mLDs can be induced to differentiate into hepatocyte-like cells.

Mouse liver ductal organoids (mLDs) cultured in Mouse Liver Ductal Organoid Expansion Medium predominantly consist of cholangiocytes. Upon switching to the Differentiation Medium, these organoids are capable of differentiating into hepatocyte-like cells, which express hepatocyte-specific markers such as Albumin, Ttr, Cyp3a11, and Mup20.

Protocol

Preparation of Mouse Liver Ductal Organoid Differentiation Medium I

Prepare Differentiation Medium I according to the table below, mix thoroughly, and keep on ice. Perform all procedures under sterile conditions.

Reagent 10 mL 20 mL 50 mL Final concentration
Mouse Liver Ductal Organoid (Expansion) Basal Medium A 9.76 mL 19.52 mL 48.8 mL
Mouse Liver Ductal Organoid (Expansion) Supplement B (50×) 200 μL 400 μL 1 mL
Mouse Liver Ductal Organoid (Expansion) Supplement C (250×) 40 μL 80 μL 200 μL

Preparation of Mouse Liver Ductal Organoid Differentiation Medium II

Reagent 10 mL 20 mL 50 mL Final concentration
Mouse Liver Ductal Organoid (Expansion) Basal Medium A 9.75 mL 19.5 mL 48.75 mL
Mouse Liver Ductal Organoid (Expansion) Supplement B (50×) 200 μL 400 μL 1 mL
Mouse Liver Ductal Organoid (Expansion) Supplement C (250×) 40 μL 80 μL 200 μL
Mouse Liver Ductal (Differentiation) Organoid Culture Supplement D (1000×) 10 μL 20 μL 50 μL

Initial Culture

Culture mouse liver ductal organoids using the Mouse Liver Ductal Organoid (Expansion) Kit (MCE Cat. No.: HY-K6118) for at least 2 days before initiating differentiation.

Differentiation Stage I

Replace the expansion medium with Differentiation Medium I and culture the mouse liver ductal organoids for 9 days. Refresh Differentiation Medium I daily during this stage.

Differentiation Stage II

Switch to Differentiation Medium II and culture for an additional 2 days. Replace Differentiation Medium II daily.

Completion of Differentiation

After Stage II, differentiation is complete and the organoids transition toward a hepatocyte-like phenotype for downstream morphology, marker, and functional analyses.
