Siglec-2/CD22

CD22 (Siglec-2) is a B cell-restricted transmembrane lectin of the Siglec family, with an extracellular V-set domain that recognizes alpha-2,6-linked sialic acids. Its cytoplasmic tail carries ITIM and ITIM-like motifs that recruit SHP-1 and restrain B cell receptor signaling, positioning CD22 as an inhibitory co-receptor. CD22 also acts as an endocytic receptor that rapidly internalizes bound antibody, a property exploited by the antibody-drug conjugate inotuzumab ozogamicin in acute lymphoblastic leukemia. Expression begins at the pre-B cell stage and persists on mature B cells and most mature B cell malignancies. Epratuzumab, an anti-CD22 antibody, has been evaluated in systemic lupus erythematosus, and Siglec ligand blockade remains an active research area.
