MCE 0.25% Trypsin-EDTA (1×), Phenol Red-Free, is prepared by dissolving trypsin powder (a mixture of proteases from porcine pancreas, irradiated and sterilized) and EDTA in a calcium- and magnesium-free balanced salt solution. It is filter-sterilized and can be directly used for the dissociation of cultured cells and tissues. The 100 mL is defined as the base specification. All larger sizes correspond to incremental volumes of this base.

Description & Advantages

Trypsin hydrolyzes intercellular proteins and disperses primary tissues or adherent cells into individual cells, and can be widely used for cell dissociation, cell culture passaging and primary tissue dissociation.

It is filter-sterilized and can be directly used for the dissociation of cultured cells and tissues.

Protocol

Dissociation of Adherent Cells

1. Completely thaw 0.25% Trypsin-EDTA (1×), no phenol red and warm to an appropriate temperature before use.

2. Remove culture medium and gently wash cells once with PBS/DPBS to remove residual serum.

3. Add enough 0.25% Trypsin-EDTA to completely cover the cell layer.

4. Incubate at 37°C for approximately 1-5 min and monitor cells microscopically. When cells round up and begin to detach, gently tap the culture vessel to facilitate detachment.

5. Add an appropriate volume of serum-containing complete medium to stop trypsin activity and gently pipette to obtain a single-cell suspension.

6. Collect the cell suspension, centrifuge if required, replace medium, and reseed according to the experimental design.

Note: Trypsin sensitivity varies among cell types. Optimize digestion time experimentally and avoid over-digestion. This is a ready-to-use, phenol-red-free 1× solution.
