Verification Images

  • Experimental Validation Results for BTF3 Antibody (YA2305)
Western blot analysis of extracts from C6 (lane 2, 20ug), NIH/3T3 (lane 3, 20ug), A549 (lane 4, 20ug) and RAW264.7 (lane 5, 20ug), Hela (lane 6, 20ug), HT-29 (lane 7, 20ug) using BTF3 Antibody (HY-P82560). Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80993, 1/10,000) was used in 5% non-fat milk in TBST for 2 hour at room temperature. Goat Anti-Rabbit IgG-HRP Secondary Antibody (HY-P8001, 1/10,000) was used for 1 hour at room temperature.
  • Experimental Validation Results for BTF3 Antibody (YA2305)
Immunohistochemical analysis of paraffin-embedded Mouse liver tissue using BTF3 Antibody (YA2305). The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P82560, 1/100) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background

  • Function

BTF3 is a When associated with NACA, prevents inappropriate targeting of non-secretory polypeptides to the endoplasmic reticulum (ER). Binds to nascent polypeptide chains as they emerge from the ribosome and blocks their interaction with the signal recognition particle (SRP), which normally targets nascent secretory peptides to the ER. BTF3 is also a general transcription factor that can form a stable complex with RNA polymerase II. Required for the initiation of transcription

  • Subcellular Localization

    Cytoplasm; Nucleus

  • Isoforms & Post-Translational Modification

    P20290 has 2 isomers: P20290-1: 22168 Da (predicted); P20290-2: 17699 Da (predicted).

  • Subunit

Part of the nascent polypeptide-associated complex (NAC), which is a heterodimer of NACA and BTF3 (via NAC-A/B domains). NAC associates with ribosomes through the BTF3/NACB subunit. Both subunits can contact nascent polypeptide chains
