  • Experimental Validation Results for HTR7 Antibody (YA2796)
    Western blot analysis of extracts from U87-MG (lane 2(20μg)), CaCo2 (lane 3(20μg)) and Rat brain (lane 4(20μg)) using 5HT7 Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (beta Actin, HY-P83730, 1/5000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Rabbit/Mouse IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

    HTR7 is a G-protein coupled receptor for 5-hydroxytryptamine (serotonin), a biogenic hormone that functions as a neurotransmitter, a hormone and a mitogen. Ligand binding causes a conformation change that triggers signaling via guanine nucleotide-binding proteins (G proteins) and modulates the activity of downstream effectors. HTR7 is coupled to G(s) G alpha proteins and mediates activation of adenylate cyclase activity[1][2].
