WB
  • Western blot analysis was performed on extracts from HepG2 (lane 1, 15 μg), U2OS (lane 2, 15 μg), RAW264.7 (lane 3, 15 μg), U-87MG (lane 4, 15 μg), HT-1080 (lane 5, 15 μg), and U251 (lane 6, 15 μg) using TRAM2 Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight. The primary antibody (1:1000 dilution) and the loading control antibody (GAPDH, HY-P80137, 1:90000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Rabbit IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.

Background
Function:E3 ubiquitin-protein ligase that regulates activation of NF-kappa-B and JNK and plays a central role in the regulation of cell survival and apoptosis (PubMed:15121867, PubMed:18997792, PubMed:19150425, PubMed:19409903, PubMed:19815541, PubMed:9390694). Catalyzes 'Lys-63'-linked ubiquitination of target proteins, such as BIRC3, IKBKE, MLST8, RIPK1 and TICAM1 (PubMed:15175328, PubMed:28489822). Is an essential constituent of several E3 ubiquitin-protein ligase complexes, where it promotes the ubiquitination of target proteins by bringing them into contact with other E3 ubiquitin ligases (By similarity). Regulates BIRC2 and BIRC3 protein levels by inhibiting their autoubiquitination and subsequent degradation; this does not depend on the TRAF2 RING-type zinc finger domain (PubMed:18997794). Plays a role in mediating activation of NF-kappa-B by EIF2AK2/PKR (By similarity). In complex with BIRC2 or BIRC3, promotes ubiquitination of IKBKE (By similarity). Acts as a regulator of mTORC1 and mTORC2 assembly by mediating 'Lys-63'-linked ubiquitination of MLST8, thereby inhibiting formation of the mTORC2 complex, while facilitating assembly of the mTORC1 complex (PubMed:28489822). Required for normal antibody isotype switching from IgM to IgG (PubMed:10514016)
Subcellular Localization:Cytoplasm
Expression:
Tissue_specificity:Isomer 1 and isomer 2 are expressed in the spleen, adipose tissue, skeletal muscle, thymus, testes, heart, lungs, and brain. Isomer 2 is expressed very weakly in the heart, lungs, and brain.
Subunit:Homotrimer (By similarity). Heterotrimer with TRAF1 (PubMed:8069916).

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
