Verification Images

  • Experimental Validation Results for ATP7B Antibody (YA2029)
Western blot analysis of extracts from HepG2(lane 2(20ug) and HepG2(lane 3(40ug) using ATP7B Antibody (HY-P82284) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P83730, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
  • Experimental Validation Results for ATP7B Antibody (YA2029)
Flow cytometric analysis of 1X106 HepG2 cells labeling ATP7B Antibody (HY-P82284, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/100 dilution for an hour at 4℃. AF488-conjugated Goat Anti-Rabbit IgG H&L (HY-P8002) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Rabbit IgG Isotype Control (HY-P80879, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
  • Experimental Validation Results for ATP7B Antibody (YA2029)
Immunocytochemistry analysis of Hela cells labeling ATP7B with ATP7B Antibody (HY-P82284) at 1/100 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with ATP7B Antibody ((HY-P82284) at 1/100 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L(HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).
  • Experimental Validation Results for ATP7B Antibody (YA2029)
Immunocytochemistry analysis of Hela cells labeling ATP7B with ATP7B Antibody (HY-P82284) at 1/50 dilution. Cells were then incubated with ATP7B Antibody ((HY-P82284) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃.

Background

  • Function

ATP7B is a Copper ion transmembrane transporter involved in the export of copper out of the cells. It is involved in copper homeostasis in the liver, where it ensures the efflux of copper from hepatocytes into the bile in response to copper overload

  • Subcellular Localization

Golgi apparatus, trans-Golgi network membrane; Multi-pass membrane protein; Late endosome; Golgi apparatus membrane; Multi-pass membrane protein; Cytoplasm; Mitochondrion

  • Expression

Tissue_specificity:Most abundant in liver and kidney and also found in brain. Isoform 2 is expressed in brain but not in liver. The cleaved form WND/140 kDa is found in liver cell lines and other tissues

  • Isoforms & Post-Translational Modification

    P35670 has 5 isomers: P35670-1: 157263 Da (predicted); P35670-2: 133617 Da (predicted); P35670-3: 145818 Da (predicted); P35670-4: 155125 Da (predicted); P35670-5: 26021 Da (predicted).
Isoform 1 may be proteolytically cleaved at the N-terminus to produce the WND/140 kDa form

  • Subunit

    Monomer. Interacts with COMMD1/MURR1 (PubMed:12968035, PubMed:17919502). Interacts with DCTN4, in a copper-dependent manner (PubMed:16554302). Interacts with ATOX1 (PubMed:17919502, PubMed:18558714). Interacts (via C-terminus) with ZBTB16/PLZF (PubMed:16676348)
