Western blot analysis of extracts from Hela (lane2(20μg), MCF-7 (lane3(20μg), A549 (lane4(20μg) and Jurkat (lane5(20μg) using PPM1D Antibody (HY-P88112). Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80993, 1/10,000) was used in 5% non-fat milk in TBST for 2 hour at room temperature. Goat Anti-Mouse IgG-HRP Secondary Antibody (HY-P8004, 1/10,000) was used for 1 hour at room temperature.
Background
-
Function
PPM1D is involved in the negative regulation of p53 expression. Required for the relief of p53-dependent checkpoint mediated cell cycle arrest. Binds to and dephosphorylates 'Ser-15' of TP53 and 'Ser-345' of CHEK1 which contributes to the functional inactivation of these proteins. Mediates MAPK14 dephosphorylation and inactivation. Is also an important regulator of global heterochromatin silencing and critical in maintaining genome integrity[1][2][3][4].

Expression
Tissue_Specificity: Expressed in fetal and adult brain. Also detected in fetal liver and skeletal muscle, but not in their adult counterparts
Induction: By p53/TP53 -
