Western blot analysis of extracts from MCF-7(lane 2(20μg) , C6 (lane 3(20μg) ,Neuro-2a(lane 4(20μg)and HEK293( lane 5(20μg) using Oct-4 Antibody（HY-P80770）.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody ( 1/1000) and Loading control antibody (Beta Actin, HY-P80993，1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (HY-P8001，1/10,000) was used for 1 hour at room temperature.

Background

Function：Transcription factor that binds to the octamer motif (5'-ATTTGCAT-3'). Forms a trimeric complex with SOX2 or SOX15 on DNA and controls the expression of a number of genes involved in embryonic development such as YES1, FGF4, UTF1 and ZFP206. Critical for early embryogenesis and for embryonic stem cell pluripotency

Expression：
Tissue_specificity:It is expressed in the developing brain. The highest expression levels are found in specific cell layers of the cerebral cortex, olfactory bulb, hippocampus, and cerebellum. Expression levels are lower in adult tissues.

Induction:Transcriptional activity is positively regulated by PKM (PubMed:18191611) . Upon cell differentiation, expression is repressed by NR6A1 (PubMed:26769970)

Sumoylation enhances the protein stability, DNA binding and transactivation activity. Sumoylation is required for enhanced YES1 expression;Ubiquitinated; undergoes 'Lys-63'-linked polyubiquitination by WWP2 leading to proteasomal degradation;ERK1/2-mediated phosphorylation at Ser-111 promotes nuclear exclusion and proteasomal degradation. Phosphorylation at Thr-235 and Ser-236 decrease DNA-binding and alters ability to activate transcription

Do most proteins show cross-species activity?
Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
