Western blot analysis was performed on extracts from HL-60 (lane 1, 15 μg), U251MG (lane 2, 15 μg), RAW264.7 (lane 3, 15 μg), and PC-12 (lane 4, 15 μg) using TIM 1 Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non - fat milk in TBST at 4°C overnight. The primary antibody (1:1000 dilution) and the loading control antibody (beta-Actin(HRP), HY-P80993, 1:5000 dilution) was incubated in 5% non-fat milk in TBST for 1 hour at 37°C. Goat Anti - Rabbit IgG - HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.

Western blot analysis of extracts from HepG2(lane 2(20ug) , Jurkat(lane 3(20ug) and NIH/3T3(lane 4(20ug) using TIM 1 Antibody (HY-P81186) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

TIM 1 is a Phosphatidylserine receptor that plays an important functional role in regulatory B-cells homeostasis including generation, expansion and suppressor functions. As P-selectin/SELPLG ligand, plays a specialized role in activated but not naive T-cell trafficking during inflammatory responses. Controls thereby T-cell accumulation in the inflamed central nervous system (CNS) and the induction of autoimmune disease. Also regulates expression of various anti-inflammatory cytokines and co-inhibitory ligands including IL10. Acts as a regulator of T-cell proliferation. May play a role in kidney injury and repair; (Microbial infection) Acts as a receptor for Hepatitis A virus; (Microbial infection) Acts as a receptor for Ebolavirus and Marburg virus by binding exposed phosphatidyl-serine at the surface of virion membrane. Serves as a dual receptor for Ebolavirus by also interacting with envelope glycoprotein GP; (Microbial infection) Acts as a receptor for Dengue virus by binding exposed phosphatidyl-serine at the surface of virion membrane. TIM1 and Dengue virus are co-internalized during virus entry; (Microbial infection) Acts as a receptor for Zika virus by binding to envelope protein E; (Microbial infection) Plays a positive role in Chikungunya virus cell entry[1][2][3][4][5][6].
