The section was pretreated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 8 minutes. Immunocytochemistry analysis of A431 cells labeling CD63 with CD63 antibody (HY-P85952) at 1/200 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 in PBS for 15 minutes at room temperature, then blocked with quick block buffer for 10 minutes at room temperature. Cells were then incubated with CD63 antibody (HY-P85952) at 1/200 dilution in quick block buffer overnight at 4 ℃. AF488-conjugated Goat Anti-Mouse IgG H&L(HY-P8005, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

Function
CD63 is a CD63, as a cell surface receptor for TIMP1, participates in the activation of the cell signaling cascade. CD63 participates in the activation of ITGB1 and integrin signals, thereby activating AKT, FAK/PTK2 and MAP kinases. CD63 promotes cell survival, actin cytoskeleton remodeling, cell adhesion, spreading and migration by activating AKT and FAK/PTK2. CD63 participates in the VEGFA signaling pathway by regulating the endocytosis of KDR/VEGFR2. CD63 participates in intracellular vesicle transport and is essential for the normal transport of the PMEL luminal domain, which is crucial for the development and maturation of melanocytes. CD63 participates in the adhesion of leukocytes to endothelial cells by regulating the transport of SELP. CD63 may play a role in mast cell degranulation in response to Ms4a2/FceRI stimulation, but does not play a role in degranulation in response to other stimuli[1].
