Western blot analysis of extracts from Hela (lane 2, 20ug), A549 (lane 3, 20ug), HL-60 (lane 4, 20ug) and K562 (lane 5, 20ug) using NEK2 Antibody (HY-P82165). Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80993, 1/10,000) was used in 5% non-fat milk in TBST for 2 hour at room temperature. Goat Anti-Rabbit IgG-HRP Secondary Antibody (HY-P8001, 1/10,000) was used for 1 hour at room temperature.

Function
NEK2 (NIMA-related kinase 2) is a serine/threonine kinase that localizes predominantly to the centrosome and functions as a key regulator of centrosome separation, bipolar spindle assembly, chromosome segregation, and mitotic progression[1][2]. NEK2 expression increases during S and G2 phases of the cell cycle, supporting its role in coordinating accurate cell division and maintenance of genomic stability[1][2]. Mechanistically, NEK2 phosphorylates proteins involved in centrosome disjunction and chromosome segregation, and dysregulated NEK2 activity promotes premature centrosome splitting, chromosomal instability, aneuploidy, and aberrant mitotic progression[1][2][3]. In addition to its canonical centrosomal functions, NEK2 can localize to the nucleus, interact with splicing factors, and regulate alternative splicing programs linked to cell survival and apoptosis[2]. In disease contexts, NEK2 is frequently overexpressed in multiple malignancies, where elevated expression is associated with tumor progression, drug resistance, chromosomal instability, and poor clinical prognosis[3][4]. Compared with other members of the NIMA-related kinase family, NEK2 is distinguished by its prominent role in centrosome dynamics and mitotic regulation, and alternative splicing generates isoforms with distinct C-terminal regions that may differ in subcellular localization and regulatory properties[1][2]. For experimental applications, NEK2 has emerged as a therapeutic target, and small-molecule inhibitors have demonstrated the ability to suppress NEK2 kinase activity in cellular and preclinical cancer models, supporting their utility for investigating mitotic regulation and cancer biology[3][5].

Expression

Induction:Expression and activity peak in the G2 phase of the mitotic cycle and decrease once the cells have entered mitosis due to degradation by the anaphase promoting complex APC/C-CDC20. In G1 phase, both isoform 1 and isoform 2 are almost undetectable. However, at the G1/S transition, there is an increase in expression of both isoforms which then remain at this increased level throughout S and G2. At the onset of mitosis, isoform 1 undergoes a rapid disappearance whereas isoform 2 continues to be present at about the same level as in G2. During the rest of mitosis, isoform 1 remains absent, while isoform 2 only begins to decline upon re-entry into the next G1 phase -
