Western blot analysis of extracts from Hela(lane 2(20ug) , MCF-7(lane 3(20ug) and HEK293(lane 4(20ug) using APAF1 Antibody (HY-P81736) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P83730, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

Immunocytochemistry analysis of Hela cells labeling APAF1 with APAF1 Antibody (HY-P81736) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with APAF1 Antibody ((HY-P81736) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L(HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

Immunohistochemical analysis of paraffin-embedded mouse heart tissue using APAF1 Antibody. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody at 1/100 dilution in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background

Function
APAF1 is an Oligomeric Apaf-1 mediates the cytochrome c-dependent autocatalytic activation of pro-caspase-9 (Apaf-3), leading to the activation of caspase-3 and apoptosis. This activation requires ATP. Isoform 6 is less effective in inducing apoptosis

Expression
Tissue_specificity:It is widely distributed. Its expression levels are highest in human spleen and peripheral blood leukocytes, and it is also expressed in the fetal brain, kidneys, and lungs. Isomer 1 is expressed in the heart, kidneys, and liver.
Induction:By E2F and p53/TP53 in apoptotic neurons (PubMed:11389439) . Translation is inhibited by HNRPA1, which binds to the IRES of APAF1 mRNAs (PubMed:31498791) -
Isoforms & Post-Translational Modification
O14727 has 6 isomers: O14727-1: 141840 Da (predicted); O14727-2: 140745 Da (predicted); O14727-3: 135980 Da (predicted); O14727-4: 137076 Da (predicted); O14727-5: 133356 Da (predicted); O14727-6: 37976 Da (predicted).

Subunit
Monomer. Oligomerizes to a heptameric ring, known as the apoptosome, upon binding of cytochrome c and dATP. Oligomeric Apaf-1 and pro-caspase-9 bind to each other via their respective NH2-terminal CARD domains and consecutively mature caspase-9 is released from the complex. Pro-caspase-3 is recruited into the Apaf-1-pro-caspase-9 complex via interaction with pro-caspase-9. Interacts with APIP. Interacts (via CARD and NACHT domains) with NAIP/BIRC1 (via NACHT domain). Interacts with CIAO2A (PubMed:25716227)
