Western blot analysis of extracts from Hela (lane2, 20μg), A549 (lane3, 20μg), PC-3 (lane4, 20μg) and MCF-7 (lane5, 20μg) using Glucose 6 phosphate isomerase Antibody (HY-P810641). Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80993, 1/10,000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (HY-P8001,1/10,000) was used for 1 hour at room temperature.

Function
Glucose 6 phosphate isomerase is an Isomerase that catalyzes the conversion of alpha-D-glucose-6-phosphate to beta-D-fructose-6-phosphate, the second step in glycolysis, and the reverse reaction in gluconeogenesis, within the cytoplasm. Also shows C2-epimerase activity, interconverting D-glucose-6-phosphate (G6P) and D-mannose-6-phosphate (M6P) (By similarity). Also displays anomerase activity, interconverting alpha and beta-anomeric forms of G6P, D-fructose-6-phosphate and M6P (By similarity). In addition to its metabolic role, this enzyme functions extracellularly as a cytokine: acts as autocrine motility factor (AMF), a secreted angiogenic factor that enhances endothelial cell motility. Functions as neuroleukin, a neurotrophic factor supporting the survival of spinal and sensory neurons. Released by lectin-stimulated T-cells to induce immunoglobulin secretion[1][2][3][4].

Isoforms & Post-Translational Modification
Glucose 6 phosphate isomerase has 2 isoforms, P06744-1: amino acid length is 558, molecular weight is 63147 Da (predicted); P06744-2: amino acid length is 569, molecular weight is 64325 Da (predicted).
Phosphorylation at Ser-185 by CK2 has been shown to decrease enzymatic activity and may contribute to secretion by a non-classical secretory pathway. -
Subunit
Homodimer; in the catalytically active form.
