Background

Function：Kinase that plays a key role in regulation of glucose and fatty acid metabolism and homeostasis via phosphorylation of the pyruvate dehydrogenase subunits PDHA1 and PDHA2 (PubMed:7499431, PubMed:18541534, PubMed:22195962, PubMed:26942675, PubMed:17683942). This inhibits pyruvate dehydrogenase activity, and thereby regulates metabolite flux through the tricarboxylic acid cycle, down-regulates aerobic respiration and inhibits the formation of acetyl-coenzyme A from pyruvate (PubMed:18541534, PubMed:22195962, PubMed:26942675). Plays an important role in cellular responses to hypoxia and is important for cell proliferation under hypoxia (PubMed:18541534, PubMed:22195962, PubMed:26942675)

Tissue_specificity:It is primarily expressed in the heart. It is also expressed in small amounts in the liver, skeletal muscle, and pancreas.

Induction:Up-regulated via the HIF1A signaling pathway in response to hypoxia

Isoforms & Post-Translational Modification：Q15118 has 2 isomers: Q15118-1: 49244 Da (predicted); Q15118-2: 51623 Da (predicted).
Phosphorylated by constitutively activated ABL1, FGFR1, FLT3 and JAK2 (in vitro), and this may also occur in cancer cells that express constitutively activated ABL1, FGFR1, FLT3 and JAK2. Phosphorylation at Tyr-243 and Tyr-244 strongly increases kinase activity, while phosphorylation at Tyr-136 has a lesser effect (PubMed:22195962). Phosphorylated under hypoxic conditions at Thr-338 by phosphoglycerate kinase PGK1 which has an activating effect (PubMed:26942675)
