Verification Images

  • Experimental Validation Results for Elk-1 Antibody (YA7598)
Western blot analysis was performed on protein extracts (30 μg) from Rat heart (lane 2), Mouse heart (lane 3), Rat brain (lane 4), and Mouse brain (lane 5) using Elk-1 antibody. Proteins were transferred onto a 0.45 μm PVDF membrane using the Trans-Blot® Turbo system for 13 min. The membrane was then blocked with 5% nonfat milk in TBST (HY-K1025) for 1 h at room temperature. Thhe primary antibody (1:2000) and loading control antibody GAPDH Antibody (HRP) (HY-P80954A) (1:5000) were diluted in 5% nonfat milk in TBST and incubated with the membrane overnight at 4°C. After washing, the membrane of primary antibody was incubated with HRP-conjugated goat anti-rabbit/mouse IgG secondary antibody (HY-P8001/HY-P8004) (1:5000) diluted in 5% nonfat milk in TBST for 1 h at room temperature. Protein bands were visualized using an Ultra High Sensitivity ECL detection kit (HY-K1005).
  • Experimental Validation Results for Elk-1 Antibody (YA7598)
Western blot analysis of extracts from HeLa (lane 1(20μg)) 、K562 (lane 2(20μg)) and Mouse brain (lane 3(20μg) using Elk-1 Antibody (HY-P87913) . Proteins were transferred to a PVDF membrane and blocked with 5% nonfat dry milk in TBST for 1.5 hour at room temperature. The primary antibody (1/4000) and Loading control antibody (β Actin, 1/10000) was used in 5% nonfat dry milk in TBST at 4℃ overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (1/10,000) was used for 1 hour at room temperature.

Background

  • Function

Elk-1 transcription factor that binds to purine-rich DNA sequences. Forms a ternary complex with SRF and the ETS and SRF motifs of the serum response element (SRE) on the promoter region of immediate early genes such as FOS and IER2. Induces target gene transcription upon JNK and MAPK-signaling pathways stimulation[1][2][3].

  • Isoforms & Post-Translational Modification

    P19419 has two isomers: P19419-1: 44888 Da (predicted); P19419-2: 11217 Da (predicted).
Sumoylation represses transcriptional activator activity as it results in recruitment of HDAC2 to target gene promoters which leads to decreased histone acetylation and reduced transactivator activity丨On mitogenic stimulation, phosphorylated on C-terminal serine and threonine residues by MAPK1

  • Subunit

Interacts in its sumoylated form with PIAS2/PIASX which enhances its transcriptional activator activity (PubMed:15920481)

  • SwissProt ID

    P19419
