  • Experimental Validation Results for Vitronectin Antibody (YA2402)
Western blot analysis was performed on extracts from 3T3 (lane 1, 15 μg), and Rat serum (lane 2, 15 μg) using Vitronectin Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight. The primary antibody (1:1000 dilution) and the loading control antibody (GAPDH, HY-P80137, 1:20000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Rabbit IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
  • Experimental Validation Results for Vitronectin Antibody (YA2402)
Immunocytochemistry analysis of HepG2 cells labeling Vitronectin Antibody (HY-P82657) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with BSA for Immunol Staining for 10 min at room temperature. Cells were then incubated with Vitronectin Antibody (HY-P82657) at 1/50 dilution in BSA for Immunol Staining at 4 ℃ Overnight. AF488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L(HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

Background

  • Function

Vitronectin is a Vitronectin is a cell adhesion and spreading factor found in serum and tissues. Vitronectin interact with glycosaminoglycans and proteoglycans. Is recognized by certain members of the integrin family and serves as a cell-to-substrate adhesion molecule. Inhibitor of the membrane-damaging effect of the terminal cytolytic complement pathway; Somatomedin-B is a growth hormone-dependent serum factor with protease-inhibiting activity

  • Subcellular Localization

    Secreted, extracellular space; Parasitophorous vacuole

  • Expression

Tissue_specificity:Expressed in the retina pigment epithelium (at protein level) (PubMed:25136834) . Expressed in plasma (at protein level) (PubMed:2448300) . Expressed in serum (at protein level) (PubMed:29567995)

  • Subunit

Exists in two forms: a single chain 75 kDa form (V75) and a clipped form composed of two chains (65 kDa and 10 kDa) (V65+V10) which are held together by a disulfide bond. Interacts with SERPINE1/PAI1, insulin and C1QBP
