Function
FOXN1 is a transcription factor that regulates the development, differentiation, and function of thymic epithelial cells (TECs) in the prenatal and postnatal thymus. As the master regulator of TEC lineage development, FOXN1 is required throughout the entire process, from the differentiation of progenitor TEC cells in the developing fetus to TEC maturation and acquisition of full function. FOXN1 directly or indirectly regulates the expression of multiple genes that mediate various aspects of thymus development and function, including MHC class II molecules, DLL4, CCL25, CTSL, CD40, and PAX1. FOXN1 regulates the differentiation of immature thymic epithelial cells (TECs) into functional cortical thymic epithelial cells (cTECs) and medullary thymic epithelial cells (mTECs). FOXN1 is crucial for maintaining the number of mTECs in the thymus after birth. It participates in the morphogenesis and maintenance of the thymic three-dimensional microstructure, which is essential for the full functioning of the thymus. It plays an important role in maintaining hematopoietic function, particularly in maintaining T cell lineage progenitor cells in the bone marrow microenvironment as the thymus ages. FOXN1 is essential for the vascularization of thymic primordia. It plays a key role in the early prenatal stages of T cell development.
