Verification Images

  • Experimental Validation Results for PKA RI alpha Antibody (YA1751)
Western blot analysis was performed on extracts from Hela (lane 1, 15 μg), Rat brain (lane 2, 15 μg), C6 (lane 3, 15 μg), Jurkat (lane 4, 15 μg), and U-87MG (lane 5, 15 μg) using PKA RI alpha Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight. The primary antibody (1:1000 dilution) and the loading control antibody (GAPDH, HY-P80137, 1:60000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Rabbit IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
  • Experimental Validation Results for PKA RI alpha Antibody (YA1751)
Western blot analysis of extracts from U87 (lane 2(20μg) and U87 (lane 3(40μg) using PKA RI alpha (HY-P82006) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

Background

  • Function

PKA RI alpha is a Regulatory subunit of the cAMP-dependent protein kinases involved in cAMP signaling in cells

  • Subcellular Localization

    Cell membrane

  • Expression

Tissue_specificity:Four types of regulatory chains are found: I-alpha, I-beta, II-alpha, and II-beta. Their expression varies among tissues and is in some cases constitutive and in others inducible

  • Isoforms & Post-Translational Modification

    P10644 has 2 isomers: P10644-1: 42982 Da (predicted); P10644-2: 38003 Da (predicted).
The pseudophosphorylation site binds to the substrate-binding region of the catalytic chain, resulting in the inhibition of its activity

  • Subunit

The inactive holoenzyme is composed of two regulatory chains and two catalytic chains. Activation by cAMP releases the two active catalytic monomers and the regulatory dimer. Interacts with PRKACA and PRKACB (PubMed:33058759).
