    Western blot analysis was performed on extracts from SH-SY5Y (lane 1, 15 μg), MDA-MB-231 (lane 2, 15 μg), Hela (lane 3, 15 μg), K562 (lane 4, 15 μg), 3T3 (lane 5, 15 μg), and C6 (lane 6, 15 μg) using DNAJB6 Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (beta-Tubulin, HY-P80955, 1:5000 dilution) was incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Rabbit IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.

    DNAJB6 is a Has a stimulatory effect on the ATPase activity of HSP70 in a dose-dependent and time-dependent manner and hence acts as a co-chaperone of HSP70. Plays an indispensable role in the organization of KRT8/KRT18 filaments. Acts as an endogenous molecular chaperone for neuronal proteins including huntingtin. Suppresses aggregation and toxicity of polyglutamine-containing, aggregation-prone proteins. Also reduces cellular toxicity and caspase-3 activity; Isoform B but not isoform A inhibits huntingtin aggregation[1][2][3][4][5].
