Western blot analysis of extracts from HepG2(lane 2(20μg), NIH-3T3(lane 3(20μg), MCF-7(lane 4(20μg) and Hela(lane 5(20μg) using PPAR alpha Antibody(HY-P86449). Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80993,1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (HY-P8001,1/10,000) was used for 1 hour at room temperature.

Function
PPAR alpha is a Ligand-activated transcription factor. Key regulator of lipid metabolism. Activated by the endogenous ligand 1-palmitoyl-2-oleoyl-sn-glycerol-3-phosphocholine (16:0/18:1-GPC). Activated by oleylethanolamide, a naturally occurring lipid that regulates satiety. Receptor for peroxisome proliferators such as hypolipidemic drugs and fatty acids. Regulates the peroxisomal beta-oxidation pathway of fatty acids. Functions as a transcription activator for the ACOX1 and P450 genes. Transactivation activity requires heterodimerization with RXRA and is antagonized by NR2C2. May be required for the propagation of clock information to metabolic pathways regulated by PER2

Isoforms & Post-Translational Modification
Q07869 has 2 isomers: Q07869-1: 52225 Da (predicted); Q07869-2: 18942 Da (predicted).
Ubiquitinated by E3 ubiquitin-protein ligase HUWE1; leading to proteasomal degradation;Phosphorylated -
Subunit
Heterodimer; with RXRA. This heterodimerization is required for DNA binding and transactivation activity. Interacts with NCOA3 coactivator. Interacts with CITED2; the interaction stimulates its transcriptional activity.
