WB
  • Western blot analysis was performed on extracts from MCF-7 (lane 1, 15 μg), HepG2 (lane 2, 15 μg), 293T (lane 3, 15 μg), and A431 (lane 4, 15 μg) using BRIP1(N-terminal) Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight. The primary antibody (1:500 dilution) and the loading control antibody (beta-Actin, HY-P80438, 1:20000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Rabbit IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.

  • Western blot analysis of extracts from Hela(lane 2(20ug) and Hela(lane 3(40ug) using BACH1 Antibody (HY-P82316) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P83730, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

Background
Function:DNA-dependent ATPase and 5'-3' DNA helicase required for the maintenance of chromosomal stability (PubMed:11301010, PubMed:14983014, PubMed:16116421, PubMed:16153896, PubMed:17596542, PubMed:36608669). Acts late in the Fanconi anemia pathway, after FANCD2 ubiquitination (PubMed:14983014, PubMed:16153896). Involved in the repair of DNA double-strand breaks by homologous recombination in a manner that depends on its association with BRCA1 (PubMed:14983014, PubMed:16153896). Involved in the repair of abasic sites at replication forks by promoting the degradation of DNA-protein cross-links: acts by catalyzing unfolding of HMCES DNA-protein cross-link via its helicase activity, exposing the underlying DNA and enabling cleavage of the DNA-protein adduct by the SPRTN metalloprotease (PubMed:16116421, PubMed:36608669). Can unwind RNA:DNA substrates (PubMed:14983014). Unwinds G-quadruplex DNA; unwinding requires a 5'-single stranded tail (PubMed:18426915, PubMed:20639400)
Subcellular Localization:Nucleus; Cytoplasm
Expression:
Tissue_specificity:It is widely expressed, with the highest expression level in the testes.
Isoforms & Post-Translational Modification:Q9BX63 has 2 isomers: Q9BX63-1: 140867 Da (predicted); Q9BX63-2: 112466 Da (predicted).
Phosphorylated. Phosphorylation is necessary for interaction with BRCA1, and is cell-cycle regulated;Acetylation at Lys-1249 facilitates DNA end processing required for repair and checkpoint signaling
Subunit:Interacts with the replication protein A complex (RPA) via the RPA1 subunit; following DNA damage they colocalize in foci in the nucleus (PubMed:17596542). Binds directly to the BRCT domains of BRCA1 (PubMed:15125843). Interacts with the CIA complex components CIAO1, CIAO2B and MMS19 (PubMed:23585563)

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
