Verification Images

  • Experimental Validation Results for Alpha B Crystallin Antibody (YA1292)
Western blot analysis was performed on protein extracts (25 μg) from Rat heart (lane 2) using Alpha B Crystallin antibody. Proteins were transferred onto a 0.45 μm PVDF membrane using the Trans-Blot® Turbo system for 13 min. The membrane was then blocked with 5% nonfat milk in TBST (HY-K1025) for 1 h at room temperature. The primary antibody (1:1000) and loading control antibody GAPDH Antibody (HRP) (HY-P80954A) (1:5000) were diluted in 5% nonfat milk in TBST and incubated with the membrane overnight at 4°C. After washing, the membrane of primary antibody was incubated with HRP-conjugated goat anti-rabbit/mouse IgG secondary antibody (HY-P8001/HY-P8004) (1:5000) diluted in 5% nonfat milk in TBST for 1 h at room temperature. Protein bands were visualized using an Ultra High Sensitivity ECL detection kit (HY-K1005).

Background

  • Function

Alpha B Crystallin may contribute to the transparency and refractive index of the lens. Has chaperone-like activity, preventing aggregation of various proteins under a wide range of stress conditions. In lens epithelial cells, stabilizes the ATP6V1A protein, preventing its degradation by the proteasome

  • Subcellular Localization

    Cytoplasm; Nucleus; Secreted; Lysosome

  • Expression

Tissue_specificity:It is expressed in the lens and other tissues (PubMed:2387586, PubMed:838078) . It is also expressed in myocardial tissue (PubMed:28493373) .
Positive sample: Rat heart tissue lysate, human skeletal muscle tissue lysate, Hela, human embryonic skeletal muscle tissue, human prostate tissue.

  • Subunit

Heteromer composed of three CRYAA and one CRYAB subunits (PubMed:20836128). Aggregates with homologous proteins, including the small heat shock protein HSPB1, to form large heteromeric complexes (PubMed:10751411). Inter-subunit bridging via zinc ions enhances stability, which is crucial as there is no protein turn over in the lens (PubMed:22890888). Interacts with HSPBAP1 and TTN/titin (PubMed:14676215). Interacts with TMEM109; in the cellular response to DNA damage (PubMed:23542032). Interacts with DES; binds rapidly during early stages of DES filament assembly and a reduced binding seen in the later stages (PubMed:28470624). Interacts with TMED10; the interaction mediates the translocation from the cytoplasm into the ERGIC (endoplasmic reticulum-Golgi intermediate compartment) and thereby secretion (PubMed:32272059). Interacts with ATP6V1A and with MTOR, forming a ternary complex (PubMed:31786107)
