Function
eIF6 (eukaryotic initiation factor 6) is a conserved ribosome biogenesis and translation factor that binds the 60S ribosomal subunit and prevents premature association with the 40S subunit, thereby regulating 80S ribosome formation and protein synthesis[1]. Mechanistically, eIF6 functions at the interface between nucleolar 60S subunit maturation and cytoplasmic translation, linking ribosome biogenesis to translational control and cellular growth[1][2]. Genetic studies demonstrated that eIF6 is required for proper 60S ribosomal subunit biogenesis, and eIF6 depletion causes a selective reduction of 60S subunits and disruption of ribosomal subunit balance[3][4]. Therefore, eIF6 acts as a critical regulator of translational capacity and growth-related protein production[4]. In disease models, eIF6 expression and activity have been associated with tumor development, and experimental reduction of eIF6 activity delays tumorigenesis and restricts tumor progression in vivo[5]. Compared with other translation initiation factors that primarily participate in initiation complex assembly, eIF6 is distinguished by its unique anti-association activity and dual role in both ribosome assembly and translation regulation[1]. Human EIF6 produces multiple transcript variants, including the predominant isoform a and a shorter isoform c generated by alternative splicing; however, current evidence primarily supports a shared role in ribosome-related functions rather than clearly distinct biological activities among isoforms[6]. For experimental applications, pharmacological or genetic inhibition of eIF6 activity has been used to suppress tumor growth and to investigate mechanisms linking ribosome biogenesis, translational control, and cancer progression[5][7].
