  • Western blot analysis of extracts from HepG2(lane 2(20ug) , Jurkat(lane 3(20ug) and MCF-7(lane 4(20ug),K562(lane 5(20ug) usin Glutamine synthetase Antibody(HY-P85817) Mouse mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P83730, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

  • Immunocytochemistry analysis of HepG2 cells labeling Glutamine synthetase with Glutamine synthetase Antibody (HY-P85817) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with BSA for Immunol Staining for 10 min at room temperature. Cells were then incubated with Glutamine synthetase Antibody (HY-P85817) at 1/50 dilution in BSA for Immunol Staining at 4 ℃, Stay overnight. AF488-conjugated AffiniPure Goat Anti-Mouse IgG H&L(HY-P8005, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

Background
Function:Glutamine synthetase that catalyzes the ATP-dependent conversion of glutamate and ammonia to glutamine (PubMed:16267323, PubMed:30158707, PubMed:36289327). Its role depends on tissue localization: in the brain, it regulates the levels of toxic ammonia and converts neurotoxic glutamate to harmless glutamine, whereas in the liver, it is one of the enzymes responsible for the removal of ammonia (By similarity). Plays a key role in ammonium detoxification during erythropoiesis: the glutamine synthetase activity is required to remove ammonium generated by porphobilinogen deaminase (HMBS) during heme biosynthesis to prevent ammonium accumulation and oxidative stress (By similarity). Essential for proliferation of fetal skin fibroblasts (PubMed:18662667). Independently of its glutamine synthetase activity, required for endothelial cell migration during vascular development: acts by regulating membrane localization and activation of the GTPase RHOJ, possibly by promoting RHOJ palmitoylation (PubMed:30158707). May act as a palmitoyltransferase for RHOJ: able to autopalmitoylate and then transfer the palmitoyl group to RHOJ (PubMed:30158707). Plays a role in ribosomal 40S subunit biogenesis (PubMed:26711351). Through the interaction with BEST2, inhibits BEST2 channel activity by affecting the gating at the aperture in the absence of intracellular L-glutamate, but sensitizes BEST2 to intracellular L-glutamate, which promotes the opening of BEST2 and thus relieves its inhibitory effect on BEST2 (PubMed:36289327)
Subcellular Localization:Cytoplasm, cytosol; Microsome; Mitochondrion; Cell membrane; Lipid-anchor
Expression:
Tissue_specificity:Expression in endothelial cells

Induction:By glucocorticoids. Vitamin D and the Wnt signaling pathway inhibit its expression and activity (PubMed:18555765) . Glutamine synthase (GS) levels are regulated by a negative feedback mechanism where elevated glutamine levels stimulate GS ubiquitin-mediated proteasomal degradation (PubMed:26990986, PubMed:38579670)
Subunit:Decamer; composed of two pentamers (PubMed:18005987). Interacts with PALMD (By similarity). Interacts with RHOJ (PubMed:30158707). Interacts with BEST2; this interaction tethers a fraction of GLUL to the membrane, causing a decrease of cytosolic glutamine synthase (GS) activity and inhibits the chloride channel activity of BEST2 by affecting the gating at the aperture in the absence of intracellular glutamate (PubMed:36289327)
Synonyms

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
