Function
GPR182 is an atypical chemokine receptor that regulates chemokine levels and localization by binding to them, without relying on the activation of classical ligand-induced signaling pathways. GPR182 mediates the sequestration, transport, or endocytosis of chemokines, thereby controlling their bioavailability. GPR182 functions as a scavenger receptor for various chemokines, including CXCL9, CXCL10, CXCL12, CXCL13, CXCL11, CXCL14, CCL1, CCL11, CCL19, CCL25, CCL28, and XCL1. GPR182 is the sole atypical receptor for the chemokines CXCL13 and CCL28. GPR182 exhibits strong constitutive binding to β-arrestin; this interaction is crucial for receptor intracellular trafficking and chemokine scavenging, and it occurs independently of ligand binding. GPR182 acts in concert with ACKR3 and ACKR4 to regulate the serum levels of CXCL12 and CCL19, respectively.
