  • Experimental Validation Results for Progesterone Receptor Antibody (YA5657)
    Western blot analysis was performed on protein extracts (40 μg) from MCF-7 (lane 2), 293T (lane 3), HeLa (lane 4) and HT-1080 (lane 5) using Progesterone Receptor antibody. Proteins were transferred onto a 0.45 μm PVDF membrane using the Trans-Blot® Turbo system for 13 min. The membrane was then blocked with 5% nonfat milk in TBST (HY-K1025) for 1 h at room temperature. The primary antibody (1:1000) and loading control antibody GAPDH Antibody (HRP) (HY-P80954A) (1:5000) were diluted in 5% nonfat milk in TBST and incubated with the membrane overnight at 4°C. After washing, the membrane of primary antibody was incubated with HRP-conjugated goat anti-rabbit/mouse IgG secondary antibody (HY-P8001/HY-P8004) (1:5000) diluted in 5% nonfat milk in TBST for 1 h at room temperature. Protein bands were visualized using an Ultra High Sensitivity ECL detection kit (HY-K1005).

    Progesterone receptor (PR) is a member of the steroid/thyroid hormone-retinoid receptor superfamily of ligand-activated nuclear transcription factors. Progesterone receptor plays a vital role in female reproductive tissue development, differentiation, and maintenance. Progesterone receptor is able to bind to a large number and variety of ligands that elicit a broad range of transcriptional responses ranging from full agonism to full antagonism and numerous mixed profiles inbetween. Progesterone receptor, such as progesterone, induces conformation changes in PR ligand binding domain (LBD), thus mediates subsequent gene regulation cascades. In humans, the biological response to progesterone is mediated by two distinct forms of the progesterone receptor (human PR-A and PR-B) .

    Tissue_specificity:In reproductive tissues, the expression of type A and type B isoforms varies with development and hormone levels. During the proliferative phase of the menstrual cycle, the expression levels of type A and type B isoforms are comparable in the uterine glandular epithelium. During the mid-secretory phase, type B isoform is persistently expressed in the glands, while type A is no longer expressed. In the stroma, type A is the predominant form throughout the cycle.
