Verification Image

Western blot analysis of extracts from THP-1(lane 2(20ug) , HL-60(lane 3(20ug) and Jurkat(lane 4(20ug) using CD161 Antibody (HY-P80966) Mouse mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P83730, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

Background

Function：Plays an inhibitory role on natural killer (NK) cells cytotoxicity. Activation results in specific acid sphingomyelinase/SMPD1 stimulation with subsequent marked elevation of intracellular ceramide. Activation also leads to AKT1/PKB and RPS6KA1/RSK1 kinases stimulation as well as markedly enhanced T-cell proliferation induced by anti-CD3. Acts as a lectin that binds to the terminal carbohydrate Gal-alpha(1,3)Gal epitope as well as to the N-acetyllactosamine epitope. Also binds to CLEC2D/LLT1 as a ligand and inhibits NK cell-mediated cytotoxicity as well as interferon-gamma secretion in target cells

Expression：
Tissue_specificity:It is primarily expressed in NK cell subsets in the intestinal epithelium and liver. It can be detected in peripheral blood T cells, especially in adult T cells with a memory antigen phenotype.

Induction:By IL12/interleukin-12 in NK cells

Subunit：Homodimer; disulfide-linked. Interacts with acid sphingomyelinase/SMPD1
