Function
Transmembrane protein 192 (TMEM192) is a lysosomal/late endosomal membrane protein containing four transmembrane segments. It exists as a homodimer, with an N-terminal dileucine motif mediating lysosomal targeting and a C-terminal cysteine motif mediating disulfide bond formation. It is widely expressed throughout the body, but is significantly overexpressed in tumor cells. Functionally, TMEM192 is involved in maintaining lysosomal membrane integrity and acidification, regulating autophagy-apoptosis crosstalk (liver cancer cells), mediating retinoic acid-induced autophagy along the TIG1-TMEM192 axis (cervical cancer), and is a key molecule in lysosomal autophagy. TMEM192 plays an important role under stress conditions such as tumor and lysosomal damage.
