  • Western blot analysis was performed on extracts from Hela (lane 1, 15 μg), A549 (lane 2, 15 μg), and MCF-7 (lane 3, 15 μg) using Cytokeratin 8 Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight. The primary antibody (1:1000 dilution) and the loading control antibody (beta-Actin, HY-P83730, 1:20000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Rabbit IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.

  • Western blot analysis of extracts from Hela(lane 2(20ug) and Jurkat(lane 3(20ug) using Cytokeratin 8 Antibody (HY-P83260) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P83730, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

  • Immunocytochemistry analysis of HeLa cells labeling Cytokeratin 8 with Cytokeratin 8 Antibody (HY-P83260) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with BSA for Immunol Staining for 10 min at room temperature. Cells were then incubated with Cytokeratin 8 Antibody (HY-P83260) at 1/50 dilution in BSA for Immunol Staining the night at 4 ℃. AF488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L(HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

Background
Function:Together with KRT19, helps to link the contractile apparatus to dystrophin at the costameres of striated muscle
Subcellular Localization:Cytoplasm; Nucleus, nucleoplasm; Nucleus matrix
Expression:
Tissue_specificity:Aggregates of dystrophin and actin are observed in muscle fibers at the sarcoplasmic sarcomere and sarcomere. They are also expressed in the gingival mucosa and hard palate of the oral cavity.
Isoforms & Post-Translational Modification:P05787 has 2 isomers: P05787-1: 53704 Da (predicted); P05787-2: 56608 Da (predicted).
Phosphorylation on serine residues is enhanced during EGF stimulation and mitosis. Ser-74 phosphorylation plays an important role in keratin filament reorganization;O-glycosylated. O-GlcNAcylation at multiple sites increases solubility, and decreases stability by inducing proteasomal degradation;O-glycosylated (O-GlcNAcylated), in a cell cycle-dependent manner
Subunit:Heterotetramer of two type I and two type II keratins (PubMed:16000376). Forms a heterodimer with KRT18 (PubMed:10852826, PubMed:24940650). Associates with KRT20 (PubMed:10973561, PubMed:16608857). Interacts with PLEC isoform 1C, when in a heterodimer with KRT18 (PubMed:24940650). Interacts with PNN (PubMed:10809736). When associated with KRT19, interacts with DMD. Interacts with TCHP (PubMed:15731013). Interacts with APEX1 (PubMed:19188445). Interacts with GPER1 (PubMed:21149639). Interacts with EPPK1 (By similarity). Interacts with PKP1 and PKP2 (PubMed:10852826)

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
