Verification Images

  • Experimental Validation Results for GNE Antibody (YA2917)
Western blot analysis of extracts from K562(lane 2(20μg) and K562(lane 3(40μg) using GNE (HY-P83172) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

Background

  • Function

GNE is a Bifunctional enzyme that possesses both UDP-N-acetylglucosamine 2-epimerase and N-acetylmannosamine kinase activities, and serves as the initiator of the biosynthetic pathway leading to the production of N-acetylneuraminic acid (NeuAc), a critical precursor in the synthesis of sialic acids. By catalyzing this pivotal and rate-limiting step in sialic acid biosynthesis, this enzyme assumes a pivotal role in governing the regulation of cell surface sialylation, playing a role in embryonic angiogenesis. Sialic acids represent a category of negatively charged sugars that reside on the surface of cells as terminal components of glycoconjugates and mediate important functions in various cellular processes, including cell adhesion, signal transduction, and cellular recognition[1][2][3][4][5][6].

  • Expression

Tissue_specificity:Highest expression in liver and placenta. Also found in heart, brain, lung, kidney, skeletal muscle and pancreas. Isoform 1 is expressed in heart, brain, kidney, liver, placenta, lung, spleen, pancreas, skeletal muscle and colon. Isoform 2 is expressed mainly in placenta, but also in brain, kidney, liver, lung, pancreas and colon. Isoform 3 is expressed at low level in kidney, liver, placenta and colon

    Q9Y223 has 5 isomers: Q9Y223-1: 79275 Da (predicted); Q9Y223-2: 83066 Da (predicted); Q9Y223-3: 78579 Da (predicted); Q9Y223-4: 71278 Da (predicted); Q9Y223-5: 66784 Da (predicted).
Phosphorylated. Phosphorylation by PKC activates the UDP-N-acetylglucosamine 2-epimerase activity

  • Subunit

Homodimer (PubMed:19841673, PubMed:22343627). Homotetramer (PubMed:26980148). Homohexamer (PubMed:19841673). The hexameric form exhibits both enzyme activities, whereas the dimeric form only catalyzes the phosphorylation of N-acyl-D-mannosamine (By similarity)
