Western blot analysis of extracts from A431(lane 2(20μg) , MCF-7(lane 3(20μg) ,SK-BR-3(lane 4(20μg)and HepG2( lane 5(20μg) using LYPD3 Antibody (HY-P81550). Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody ( 1/1000) and Loading control antibody (Beta Actin, HY-P80993,1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (HY-P8001,1/10,000) was used for 1 hour at room temperature.

Immunohistochemical analysis of paraffin-embedded human cervical cancer tissue using LYPD3 Antibody (YA1295). The section was pre-treated using heat mediated antigen retrieval with Tris/EDTA buffer (pH 9.0) for 20 minutes. The tissues were probed with the primary antibody (HY-P81550, 1/100) overnight at 4℃. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with neutral balsam.

Background
-
Function
LYPD3 supports cell migration. May be involved in urothelial cell-matrix interactions. May be involved in tumor progression
-
Subcellular Localization
Cell membrane; Lipid-anchor, GPI-anchor
-
Expression
Tissue_specificity:It is expressed in the placenta, skin, and urothelial cells. It is found in keratinocytes in the basal layer of chronic wounds. It is weakly expressed in esophageal and peripheral blood mononuclear cells. It is found in most primary and metastatic transitional cell carcinomas (TCCs) and breast cancer tissues, but not in adjacent normal tissues. High expression is found in tumor components of some non-invasive superficial lesions, as well as in invasive and metastatic urothelial carcinomas.
Induction:Up-regulated in migrating keratinocytes during epithelization of incisional skin wounds -
Subunit
Binds laminin-1 and laminin-5. Interacts with LGALS3 (By similarity). Interacts with AGR2 and AGR3
