  • Function

    Repulsive guidance molecule A (RGMa) is a GPI-anchored glycoprotein that exhibits repulsive neurite guidance and regulates neuronal differentiation and survival during brain development[1]. Mechanistically, RGMa signals through neogenin and Rho-associated protein kinase, suppressing neurite outgrowth and adult neurogenesis in the dentate gyrus[1][2]. In neural circuit models, RGMa repels entorhinal axons and supports layer-specific termination of the perforant pathway[3]. In disease models, RGMa overexpression induces dopaminergic neuron degeneration and Parkinson’s disease-like behavioral and anatomical changes[4]. Compared with related isoforms, RGMa, RGMb, and RGMc are distinct single-copy genes with discrete expression patterns and unique biological functions[5]. RGM family proteins bind selected BMPs, but RGMa shows lower affinity for most tested BMPs than RGMB or HJV/RGMc[6]. For experimental applications, RGMa blockade increased afferent synapse formation with auditory hair cells, while recombinant RGMa suppressed seizures and mossy fiber sprouting in a PTZ kindling model[7][8].
