Western blot analysis of extracts from Mouse spleen tissue (lane2, 20μg), NIH/3T3 (lane3, 20μg), and RAW264.7 (lane4, 20μg) using Caspase-8 (p18) Antibody (HY-P86835). Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/2000) and Loading control antibody (Beta Actin, HY-P80993, 1/10,000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (HY-P8001,1/10,000) was used for 1 hour at room temperature.

Immunocytochemistry analysis of Hela cells labeling Caspase-8 (p18) With Caspase-8 (p18) antibody (HY-P86835) at 1/100 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 in PBS for 15 minutes at room temperature, then blocked with quick block buffer for 10 minutes at room temperature. Cells were then incubated with Caspase-8 (p18) antibody (HY-P86835) at 1/100 dilution in quick block buffer overnight at 4 ℃. AF488-conjugated Goat Anti-Rabbit IgG H&L(HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

Function
Caspase 8 is an initiator protease of death receptor-mediated extrinsic apoptosis and also supports cell survival by restraining RIP-family kinase-dependent necroptosis[1]. Mechanistically, caspase 8 works with cFLIP to regulate the switch among survival, apoptosis, and necroptosis, while cFLIP is structurally related to caspase 8 but lacks proteolytic activity[1][2]. Therefore, caspase 8 differs from inflammatory caspases such as caspase 1, 4, 5, and 12, which participate in inflammasome-related innate immune responses[3]. In intestinal disease, caspase 8 controls Paneth cell necroptosis and may contribute to epithelial cell death and mucosal inflammation in Crohn’s disease[3]. In severe SARS-CoV-2 mouse models, non-apoptotic caspase 8 promoted cytokine release, IL-1β elevation, NF-κB-linked inflammation, and disease pathology[4]. In ovarian cancer, low caspase 8 expression correlated with shorter overall survival, while caspase 8 loss enhanced non-apoptotic necroptotic death under NF-κB blockade[5]. For experimental applications, studies use genetic caspase 8 loss, viral caspase 8 inhibitors, and necroptosis pathway modulation to separate apoptosis, inflammation, and RIPK3-MLKL-dependent cell death[6].
