Western blot analysis of extracts from A549(lane 2, 20μg), A431(lane 3, 20μg), HeLa(lane 4, 20μg) and HepG2(lane 5, 20μg) using EPHX1 Antibody (HY-P88203). Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hours at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80993, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse IgG-HRP Secondary Antibody (HY-P8004, 1/10000) was used for 1 hour at room temperature.

Flow cytometric analysis of 1X106 HepG2 cells labeling EPHX1 Antibody (HY-P88203, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/100 dilution for an hour at 4℃. AF488-conjugated Goat Anti-Mouse IgG H&L (HY-P8005) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Mouse IgG Isotype Control (HY-P80757, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).

EPHX1 is a Biotransformation enzyme that catalyzes the hydrolysis of arene and aliphatic epoxides to less reactive and more water soluble dihydrodiols by the trans addition of water. Plays a role in the metabolism of endogenous lipids such as epoxide-containing fatty acids. Metabolizes the abundant endocannabinoid 2-arachidonoylglycerol (2-AG) to free arachidonic acid (AA) and glycerol. Binds 20(S)-hydroxycholesterol (20(S)-OHC)[1][2].
