Western blot analysis of extracts from Daudi (lane2, 20μg), Raji (lane3, 20μg), LOVO (lane4, 20μg) and Jurkat (lane5, 20μg) using CD74 Antibody (HY-P85898). Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80993, 1/10,000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse IgG-HRP Secondary Antibody (HY-P8004,1/10,000) was used for 1 hour at room temperature.

Function
CD74 (HLA class II histocompatibility antigen gamma chain; HLA-DR antigens-associated invariant chain) is a type II transmembrane protein. CD74 is involved in the formation and transport of MHC class II peptide complexes for the generation of CD4+ T cell responses. Cell surface CD74 serves as a receptor for the cytokine, macrophage migration inhibitory factor (MIF) on many cell types. In immune cells, MIF binding to CD74 induces a signaling cascade that results in regulation of cell proliferation and survival. CD74 has been associated with tumor progression and metastasis. CD74 expression has been suggested to serve as a prognostic factor in many cancers, with higher relative expression behaving as a marker of tumor progression[1].

Subunit
Homotrimer. In the endoplasmic reticulum (ER) it forms a heterononameric MHC II-Ii complex: 3 MHC class II molecules (heterodimers of an alpha and a beta subunit) bind to the CD74 homotrimer (also known as invariant chain or HLA class II histocompatibility antigen gamma chain). In the endosomal/lysosomal system, the CD74 component undergoes sequential degradation by various proteases, including CTSS and CTSL, leaving a small fragment termed CLIP (class-II-associated invariant chain peptide) attached to the MHC class II molecule (alpha-beta-CLIP complex). This processed complex interacts with HLA_DM and HLA_DO heterodimers in order to release CLIP and facilitate the binding of antigenic peptides to the MHC class II molecules. Interacts with CD44; this complex is essential for the MIF-induced signaling cascade that results in B cell survival
