  • Experimental Validation Results for CD20 Antibody (YA6136)
Western blot analysis of extracts from Hela (lane2(20μg), Raji (lane3(20μg), Ramos (lane4(20μg) and Daudi (lane5(20μg) using CD20 Antibody (HY-P86444). Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/3000) and Loading control antibody (Beta Actin, HY-P80993, 1/10,000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (HY-P8001 ,1/10,000) was used for 1 hour at room temperature.
  • Experimental Validation Results for CD20 Antibody (YA6136)
Immunohistochemical analysis of paraffin-embedded human tonsil tissue using CD20 Antibody (HY-P86444, 1/2000). The section was pretreated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 8 minutes. The tissues were blocked with quick block buffer for 0.5 hours at room temperature, washed with PBS and PBST, and then incubated with the primary antibody overnight at 4℃. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background

  • Function

CD20 is a 33-37 kDa non-glycosylated protein expressed specifically on the surface of normal and malignant B lymphocytes, and belongs to the MS4A (membrane-spanning 4-domain family A) protein family. CD20 is a general B-cell marker expressed by the majority of B cells starting from late pre-B lymphocytes (it is not expressed by pro-B lymphocytes), and its expression is lost in terminally differentiated plasmablasts and plasma cells. Early experiments suggested that it was important in the regulation of human B-cell activation, proliferation, and Ca2+ transport. However, CD20-deficient mice appeared normal. Hence, the exact biological function of CD20 and its physiological ligand remain to be fully defined.240 Regardless of the uncertainty in the biological role of this antigen, it is a good target for cancer immunotherapy.
