    Hydroxyacid oxidase 1 (HAO1) functions as a rate-limiting enzyme in oxalate biosynthesis, catalyzing glycolate oxidation to glyoxylate, which is subsequently converted to oxalate[1]. Mechanistically, HAO1 activity in alveolar epithelial cells triggers oxalate accumulation, which activates NADPH oxidase and induces neutrophil extracellular trap (NET) formation, promoting pre-metastatic niche establishment in the lung[1]. This HAO1-mediated oxalate accumulation also enhances metastatic breast cancer cell proliferation via MAPK signaling activation[1]. Compared with related oxidase isoforms, HAO1 exhibits tissue-specific upregulation in pulmonary alveolar epithelium during the pre-metastatic stage, distinct from baseline hepatic expression of other hydroxyacid oxidases[1].
