Function
Slit-2 (SLIT2) is a secreted ligand that binds ROBO receptors and functions as a repulsive axon-guidance cue[1]. Mechanistically, SLIT2/ROBO1 signaling regulates cytoskeletal remodeling, inhibits macrophage macropinocytosis, and limits NOD2-NF-κB-CXCL1 inflammatory signaling[2]. In immune models, Slit inhibited leukocyte chemotaxis, while Slit2 administration reduced neutrophil infiltration, tubular injury, and renal failure after renal ischemia-reperfusion injury[3][4]. In cancer models, SLIT2 biology is context-dependent: Slit-Robo signaling promoted tumor angiogenesis in melanoma, whereas SLIT2 inactivation was reported in lung and breast cancers[5][6]. Compared with related isoforms, SLIT1 is mainly nervous-system-associated, while SLIT2 and, to a lesser extent, SLIT3 are also detected in peripheral tissues[2]. For experimental applications, recombinant N-terminal SLIT2 (NSlit2) activates ROBO-dependent signaling, while ROBO1 neutralization or SLIT2-trap strategies block pathway activity in angiogenesis and glioblastoma models[2][5][7].
