Verification Images

  • Experimental Validation Results for ORP150 Antibody (YA1478)
Western blot analysis of extracts from MCF-7 (lane 2(20μg), MCF-7 (lane 3(40μg), using ORP150 Antibody. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.
  • Experimental Validation Results for ORP150 Antibody (YA1478)
Immunohistochemical analysis of paraffin-embedded rat testis tissue using ORP150 Antibody. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 2 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody at 1/100 dilution in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background

  • Function

ORP150 is a Has a pivotal role in cytoprotective cellular mechanisms triggered by oxygen deprivation. Promotes HSPA5/BiP-mediated ATP nucleotide exchange and thereby activates the unfolded protein response (UPR) pathway in the presence of endoplasmic reticulum stress. May play a role as a molecular chaperone and participate in protein folding

  • Subcellular Localization

    Endoplasmic reticulum lumen

  • Expression

Tissue_specificity:Highly expressed in tissues that contain well-developed endoplasmic reticulum and synthesize large amounts of secretory proteins. Highly expressed in liver and pancreas and lower expression in brain and kidney. Also expressed in macrophages within aortic atherosclerotic plaques, and in breast cancers

Induction:By hypoxia and also by 2-deoxyglucose or tunicamycin

  • Isoforms & Post-Translational Modification

    Q9Y4L1 has 2 isomers: Q9Y4L1-1: 111335 Da (predicted); Q9Y4L1-2: 62757 Da (predicted).

  • Subunit

Part of a large chaperone multiprotein complex comprising DNAJB11, HSP90B1, HSPA5, HYOU, PDIA2, PDIA4, PDIA6, PPIB, SDF2L1, UGGT1 and very small amounts of ERP29, but not, or at very low levels, CALR nor CANX
