Western blot analysis of extracts from HEK293(lane 2(20ug) , Hela(lane 3(20ug) and Jurkat(lane 4(20ug) using HtrA2 Antibody (HY-P80715) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
Background

Function
HtrA2/Omi serine protease that shows proteolytic activity against a non-specific substrate beta-casein. Promotes apoptosis by either relieving the inhibition of BIRC proteins on caspases, leading to an increase in caspase activity; or by a BIRC inhibition-independent, caspase-independent and serine protease activity-dependent mechanism. Cleaves BIRC6 and relieves its inhibition on CASP3, CASP7 and CASP9, but it is also prone to inhibition by BIRC6. Cleaves THAP5 and promotes its degradation during apoptosis; Seems to be proteolytically inactive[1][2][3][4][5].

O43464 has 4 isomers: O43464-1: 48841 Da (predicted); O43464-2: 38493 Da (predicted); O43464-3: 46382 Da (predicted); O43464-4: 39914 Da (predicted).
Ubiquitinated by BIRC6; this activity is inhibited by DIABLO/SMAC;Autoproteolytically activated -
Subunit
Homotrimer (PubMed:36758104, PubMed:36758105). Interacts with MXI2. Interacts with THAP5 under apoptotic conditions. The mature protein, but not the precursor, binds to BIRC2/c-IAP1, BIRC3/c-IAP2 and XIAP/BIRC4. Interacts with AREL1 (via HECT domain); in the cytoplasm following induction of apoptosis (PubMed:23479728)
