Background
-
Function
ETS1 transcription factor. Directly controls the expression of cytokine and chemokine genes in a wide variety of different cellular contexts. May control the differentiation, survival and proliferation of lymphoid cells. May also regulate angiogenesis through regulation of expression of genes controlling endothelial cell migration and invasion; Acts as a dominant-negative for isoform c-ETS-1A[1][2][3][4][5].

Expression
Tissue_specificity:It is highly expressed in lymphocytes. Both c-ETS-1A and Ets-1 p27 subtypes were detected in all fetal tissues tested, but in human tissues, the variation varied depending on the tissue type. It was not detected in brain tissue or kidneys.
Induction:Up-regulated by retinoic acid, VEGF, TNF-alpha/TNFA, lipopolysaccharide and in response to hypoxia (at protein level) -
Isoforms & Post-Translational Modification
P14921 has 5 isomers: P14921-1: 50408 Da (predicted); P14921-2: 40786 Da (predicted); P14921-3: 55412 Da (predicted); P14921-4: 25697 Da (predicted); P14921-5: 30860 Da (predicted).
Sumoylated on Lys-15 and Lys-227, preferentially with SUMO2; which inhibits transcriptional activity;Ubiquitinated; which induces proteasomal degradation;Phosphorylation at Ser-251, Ser-282 and Ser-285 by CaMK2/CaMKII in response to calcium signaling decreases affinity for DNA: an increasing number of phosphoserines causes DNA-binding to become progressively weaker -
Subunit
Binds DNA as a homodimer; homodimerization is required for transcription activation (PubMed:18566588). Interacts with MAF and MAFB (By similarity). Interacts with PAX5; the interaction alters DNA-binding properties (By similarity). Interacts with DAXX (PubMed:10698492). Interacts with UBE2I (PubMed:9333025). Interacts with SP100; the interaction is direct and modulates ETS1 transcriptional activity (PubMed:11909962, PubMed:15247905)
