  • Experimental Validation Results for CD8 alpha Antibody (YA6308)
Western blot analysis of extracts from MCF-7 (lane2(20μg), THP-1(lane3(20μg), Jurkat(lane4(20μg) and Raji(lane5(20μg) using CD8 alpha Antibody (YA6308) (HY-P86616). Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight. The primary antibody (1/2000) and Loading control antibody (Beta Actin, HY-P80993, 1/10,000) was used in 5% non-fat milk in TBST for 2 hour at room temperature. Goat Anti-Rabbit IgG-HRP Secondary Antibody (HY-P8001, 1/10,000) was used for 1 hour at room temperature.
  • Experimental Validation Results for CD8 alpha Antibody (YA6308)
Immunohistochemical analysis of paraffin-embedded human spleen tissue using CD8 alpha Antibody (HY-P86616, 1/1000). The section was pretreated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 8 minutes. The tissues were blocked with quick block buffer for 0.5 hours at room temperature, washed with PBS and PBST, and then incubated with the primary antibody overnight at 4℃. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background

  • Function

CD8a (CD8α) is a key component of the CD8 co-receptor and is primarily expressed on cytotoxic CD8+ T cells. By enhancing TCR-MHC I signaling, it promotes T-cell activation, IFN-γ production, and granzyme/perforin-mediated cell killing; it is one of the most commonly used T-cell markers in tumor immunology, infection immunology, vaccine research, and CAR-T cell research.

  • Subcellular Localization

Cell membrane; Single-pass type I membrane protein; Secreted

  • Expression

Tissue_specificity:CD8 on thymus-derived T-cells usually consists of a disulfide-linked alpha/CD8A and a beta/CD8B chain. Less frequently, CD8 can be expressed as a CD8A homodimer. A subset of natural killer cells, memory T-cells, intraepithelial lymphocytes, monocytes and dendritic cells expresses CD8A homodimers. Expressed at the cell surface of plasmacytoid dendritic cells upon herpes simplex virus-1 stimulation

  • Isoforms & Post-Translational Modification

    CD8a has 3 isomers: P01732-1: 25729 Da (predicted); P01732-2: 21585 Da (predicted); P01732-3: 30214 Da (predicted).
Palmitoylated, but association with CD8B seems to be more important for the enrichment of CD8A in lipid rafts;O-glycosylated;Phosphorylated in cytotoxic T-lymphocytes (CTLs) following activation

  • Subunit

Forms disulfide-linked heterodimers with CD8B at the cell surface. Also forms homodimers in several cell types including NK-cells or peripheral blood T-lymphocytes.
