    Western blot analysis of extracts from K562 (lane 2(20μg), K562 (lane 3(40μg), using NFAT2 Antibody. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.

    Flow cytometric analysis of 1X10^6 K562 cells labeling NFAT2 Antibody (HY-P80243, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/1000 dilution for an hour at 4℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L (HY-P8002) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Rabbit IgG Isotype Control (HY-P80879, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).

    Plays a role in the inducible expression of cytokine genes in T-cells, especially in the induction of the IL-2 or IL-4 gene transcription. Also controls gene expression in embryonic cardiac cells. Could regulate not only the activation and proliferation but also the differentiation and programmed death of T-lymphocytes as well as lymphoid and non-lymphoid cells (PubMed:10358178).     Tissue_specificity:This protein is expressed in the thymus, peripheral blood leukocytes (such as T cells) , and spleen. The A subtype is preferentially expressed in effector T cells (thymus and peripheral blood leukocytes) , while the B and C subtypes are preferentially expressed in naive T cells (spleen) . The B subtype is expressed after the first exposure of naive T cells to the antigen, and the A subtype is expressed after the second exposure of effector T cells to the antigen. The IA subtype is widely expressed but not detected in the liver and pancreas; its neural expression is strongest in the corpus callosum. The IB subtype is mainly expressed in muscle, cerebellum, placenta, and thymus; neural expression is present in all of these areas of the human brain, with the strongest expression in the corpus callosum.
